EGFRdimerizationstimulatesitsintrinsicintracellularprotein-tyrosinekinaseactivity.Asaresult,autophosphorylationoffivetyrosine(Y)residuesintheC-terminaldomainofEGFRoccurs.TheseareY992,Y1045,Y1068,Y1148andY1173.Thisautophosphorylationelicitsdownstreamactivationandsignalingbyseveralotherproteinsthatassociatewiththephosphorylatedtyrosinesthroughtheirownphosphotyrosine-bindingSH2domains.Thesedownstreamsignalingproteinsinitiateseveralsignaltransductioncascades,principallytheMAPK,AktandJNKpathways,leADIngtoDNAsynthesisandcellproliferation.Suchproteinsmodulatephenotypessuchascellmigration,adhesion,andproliferation.ThekinasedomainofEGFRcanalsocross-phosphorylatetyrosineresiduesofotherreceptorsitisaggregatedwith,andcanitselfbeactivatedinthatmanner. |
