ImmunohistochemicallocalizationofCNPasehasshowntheenzymetoberestrictedtooligodendrocytesandSchwanncells.Theenzymeappearstobedistributedinsingleanddoubleloosewrapsofmyelinandnotincompactmyelinasearlierthoughtbymostinvestigators.CNPaseislocatedontheinnerandouterloopsofmyelin,paranodallyandneartheinnersurfaceoftheoligodendrocytemembrane.Inplaqueregionsofmultiplesclerosispatients,theenzymeisreduced,andwhenCNSmyelinisdecreased,CNPaseisoneoftheearlierproteinstobelostfromthemyelin.Inaddition,anenzymethatisprobablyidenticaltobrainCNPaseislocatedintheouterrodsegmentswithinthevisualsystem,andthisproteinisalsorecognizedbythemonoclonalantibody11-5B.Inmixedhumangliomas,theenzymelevelsarereduced,althoughabout5%oftheoligodendrocytesoccasionallyshownormalpositivestaining. |
