BackgroundInformationHistonesarehighlyconservedproteinsthatserveasthestructuralscaffoldfortheorganizationofnuclearDNAintochromatin.Thefourcorehistones,H2A,H2B,H3,andH4,assembleintoanoctamer(2moleculesofeach).Histonesaremodifiedpost-translationallybytheactionsofenzymesinboththenucleusandcytoplasm.Thesemodifications,whichincludeacetylation,phosphorylation,methylation,andubiquitin-ation,occurpredominantlyontheN-terminalandC-terminaltailsthatextendbeyondthenucleosomecoreparticle.Argininemodificationsofhistonesproteinsaremediatedbytwoclassesofproteinargininemethyltransferases(PRMTs),classIandII.ClassIPRMTsplacetwomethylgroupsontheonenitrogenatomofthearginineguanidinogroup,resultinginanasymmetricmodification.IncontrastclassIIPRMTsplacetwomethylgroupsontwonitrogenatomsoftheguanidinogroupproducingasymmetricdimethylation.Althoughthereareexceptions,asymmetricmodificationsaretypicallyassociatedwithtranscriptionactivationwhilesymmetricmodificationsaremostcommonlyassociatedwithtranscriptionalrepression.
