Earlytechniqueswerebasedontheperoxidase-anti-peroxidase(PAP)reaction28,whileimprovementsexploitedthestrongaffinityofavidinforbiotin10.ThistechniqueusesHRP-labeledstreptavidin.Thesecondaryantibodyisconjugatedwithbiotinandthestreptavidin-HRPcomplexreactswiththebiotinonthesecondaryantibodies.Theresultingbiotin-avidin-HRPcomplexcanreactwiththeprimaryantibodyboundtothespecificepitopeofthetargetantigen.TheHRPenzymesofthestreptavidincomplexthencatalyzethesubstrate/chromogenreactiontoformacoloredreactionproduct(browntoblackwhenusingDABasthechromogen)attheantigensite.ABIOLOGicalstainisthenusedtovisualizethewholetissuesection. |
