| Overview | SummaryandExplanation: Immunohistochemicaltechniqueshavebeeninuseasadjunctivemethodsfortherecognitionofcellsintissuesectionsforatleast50years.Thefirstmethodreporteduseoffluorescentlabelsin19421.In1974,reportsofusingenzymaticlabels,suchashorserADIshperoxidase(HRP)inroutineH&Eparaffinsectionsappeared2,3.Themethodshavesincebecomethe"standardofcare"insurgicalpathologywhenclassicmethodsalonefailtoyieldadefinitivediagnosis4-9. Earlytechniqueswerebasedontheperoxidase-anti-peroxidase(PAP)reaction28,whileimprovementsexploitedthestrongaffinityofavidinforbiotin10.ThistechniqueusesHRP-labeledstreptavidin.Thesecondaryantibodyisconjugatedwithbiotinandthestreptavidin-HRPcomplexreactswiththebiotinonthesecondaryantibodies.Theresultingbiotin-avidin-HRPcomplexcanreactwiththeprimaryantibodyboundtothespecificepitopeofthetargetantigen.TheHRPenzymesofthestreptavidincomplexthencatalyzethesubstrate/chromogenreactiontoformacoloredreactionproduct(browntoblackwhenusingDABasthechromogen)attheantigensite.ABIOLOGicalstainisthenusedtovisualizethewholetissuesection. |
