| Overview | Caspase-2(alsoknownasICH-1,NEDD-2)isamemberoftheinterleukin-1betaconvertingenzyme(ICE)familyofcysteinproteases.Similarasothercaspases,caspase-2alsoexistsincellsasaninactiveproenzyme.Duringapoptosis,procaspase-2isprocessedataspartateresiduesbyself-proteolysisand/orcleavagebyupstreamcaspases.Theprocessedformofcaspase-2consistsoflarge(19kDa)andsmall(12kDa)subunitswhichassociatetoformtheactiveenzyme.Theactiverecombinanthumancaspase-2wasexpressedinE.coli.Theexpressedcaspase-2spontaneouslyundergoesautoprocessingtoyieldthesubunitscharacteristicofthenativeenzyme.Therecombinantcaspase-2preferentiallycleavesthesubstratesconsistingofconsensussequenceVDVAD(e.g.,VDVAD-AFCandVDVAD-pNA).Therecombinantenzymecanalsobeusedasapositivecontrolincaspaseassaysorindeterminingthespecificityofcaspasesubstrates.Recommendedusage:0.5-1unitforfluorometriccaspaseassaysand2-4unitsforcolorimetriccaspaseassays.Optimalworkingdilutionsmustbedeterminedbyenduser. |
