Theactiverecombinanthumancaspase-1wasexpressedinE.coli.Theexpressedcaspase-1spontaneouslyundergoesautoprocessingtoyieldthesubunitscharacteristicofthenativeenzyme.Therecombinantcaspase-1preferentiallycleavesthesubstratesconsistingofconsensussequenceYVAD(e.g.,YVAD-AFCandYVAD-pNA).Therecombinantenzymecanalsobeusedasapositivecontrolincaspaseassaysorindeterminingthespecificityofcaspasesubstrates.
