TheabilitytodetectthephosphorylationofEGFRandERKsimultaneouslyallowsresearcherstoprofileEGFmediatedMAPKactivationwithinacelltypeofinterest.Alterationsinthesesignalingprofilescanbeusedtomonitortheeffectofaparticularupstreamtreatmenttargetingthepathway. Antibodiesagainstphospho-EGFRandphospho-ERK1/2areprovidedinthekittomeasuretheextentofeachproteinsactivationstate.A431cellswereusedasamodelsystemtodemonstrateEGFmediatedactivationofEGFRaswellasits’downstreameffectorkinaseERK.Thetreatedcellswerecomparedtountreatedcellstoverifythedetectionofbothactivatedproteinsbyflowcytometry.Whenperformingadualcolorstain,themajorityofthetreatedcellsaredoublepositiveindicatingactivationofMAPKthroughEGFR.
